They are among five players who played in the Copa America earlier this summer and have yet to begin training ahead of the upcoming season.
Claudio Bravo, Javier Mascherano, Dani Alves will also skip the three-match International Champions Cup, which starts against the LA Galaxy at the Rose Bowl on Tuesday, followed by games against Premier League clubs Manchester United and Chelsea.
It is the second time this year that American fans will miss the chance to watch Messi, who also sat out two Argentina friendlies in March with a foot injury.
New Barcelona signings Arda Turan and Aleix Vidal will both be on the 24-man squad.
